Synthesis rates of myosin and actin in skeletal muscle of critically ill patients

چکیده
Introduction Critical illness is characterized by a dramatic loss of skeletal muscle mass which comprises to about 10% per week. This loss is mostly driven by an increased muscle protein breakdown, whereas synthesis rates are on average not changed [1]. However, this result limits to total muscle protein and does not elucidate whether different proteins behave differently. We have previously shown that mitochondrial protein synthesis in skeletal muscle of critically ill patients is not different from controls [2].

Isolation of human skeletal muscle myosin heavy chain and actin for measurement of fractional synthesis rates.
Using sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), we have developed a simple method to isolate myosin heavy chain (MHC) and actin from small (60-80 mg) human skeletal muscle samples for the determination of their fractional synthesis rates. The amounts of MHC and actin isolated are adequate for the quantification of [13C]leucine abundance by gas chromatography-combusti...
متن کاملThe Effect of Resistance Training and Endothelial Stem Cell Injection on Βeta-Actin, Phosphorylated and Total AKT of Skeletal Muscle in Type 1 Diabetic Rats
Background: Type 1 diabetes is associated with decreased skeletal muscle capillary and improper regulation of angiogenesis pathways in skeletal muscle. This research intended to study the effect of resistance training and endothelial stem cell injection on βeta-actin, phosphorylated and total AKT of skeletal muscle in type 1 diabetic rats. Methods: In this experimental study, 36 male Wistar ra...

New Zealand White rabbits were infused with [3H]tyrosine for periods of 5--6 h and then different methods of extraction were applied for the purification of the main muscle proteins and protein fractions. Myosin (I), prepared from salt extraction of muscle mince, consistently had a higher specific radioactivity than did myosin (II), isolated by dissociation of actomyosin. Actins (IA) and (IB), ...
متن کاملNutritional Assessment in Critically Ill Patients
Background: Malnutrition is an important factor in the survival of critically ill patients. The purpose of the present study was to assess the nutritional status of patients in the intensive care unit (ICU) on the days of admission and discharge via a detailed nutritional assessment. Methods: Totally, 125 patients were followed up from admission to discharge at 8ICUs in Shiraz, Iran. The patien...
متن کاملProtein metabolism and gene expression in skeletal muscle of critically ill patients with sepsis.
Muscle wasting negatively affects morbidity and mortality in critically ill patients. This progressive wasting is accompanied by, in general, a normal muscle PS (protein synthesis) rate. In the present study, we investigated whether muscle protein degradation is increased in critically ill patients with sepsis and which proteolytic enzyme systems are involved in this degradation. Eight patients...
